Channel catfish virus (CCV) causes an acute haemorrhagic disease in channel catfish, Ictalurus punctatus (Rafinesque), fry and fingerlings. The present study describes a polymerase chain reaction (PCR)-based assay for detection of CCV DNA in the tissues of acutely infected juvenile catfish. The assay is rapid, sensitive and specifically detects CCV DNA derived from epidemiologically distinct viral isolates. The use of two independent PCR primers sets, each specific for particular CCV genes (open reading frames 8 and 59), provides a means to confirm the results and minimize false-positive results. The method identifies CCV DNA in several tissues of acutely infected fish, including the brain, blood, intestine, kidney and liver. The CCV PCR assay is useful for the diagnosis of acute CCV disease and for studies to investigate the molecular basis of CCV pathogenesis. 相似文献

We investigated the in vitro effect of dibenzyl trisulfide (DTS), a secondary metabolite of Petiveria alliacea, on erythrocyte elasticity, relaxation time and membrane morphology. Blood samples from 8 volunteers with hemoglobin AA were exposed to 100, 200, 400, 800 and 1000 ng/ml of DTS respectively and the elasticity and relaxation time measured. There were statistically significant, dose-dependent increases in elasticity and relaxation times. The changes in membrane morphology observed also increased with increased concentration of DTS. This suggests that DTS interaction with membrane protein resulted in increased elasticity, relaxation time and deformation of the erythrocyte membrane. 相似文献

Methicillin‐resistant Staphylococcus aureus (MRSA) is an emerging cause of serious bacterial infection in the horse, with an increasing number of cases reported over the last decade. MRSA, along with other commensal staphylococcal species, can reside on the mucosa of several sites in the horse, particularly the nose. Nasal carriage of MRSA appears rare amongst horses in the community, although a higher prevalence has been found in hospitalised horses. MRSA infections can involve a variety of body sites, but most commonly encountered are soft tissue infections of either traumatic or surgical wounds. MRSA strain types isolated from horses are typically multidrug‐resistant and usually differ from those recovered from humans and other small animal species. Treatment of infection can be prolonged and is dependent on timely, accurate diagnosis and on appropriate therapy; often guided by antimicrobial susceptibility testing. The purpose of this review is to provide clinically relevant information for the equine practitioner and, for illustration, the diagnosis, treatment and outcome of 4 clinical cases of MRSA infection in horses is discussed. 相似文献

Staphylococcus pseudintermedius is part of the normal canine flora but frequently causes pyoderma in canine atopic dermatitis (AD). This study aimed to determine whether particular S. pseudintermedius strains were associated with AD and/or pyoderma. Ninety‐six S. pseudintermedius isolates from the ear, nares, perineum and lesions of 21 atopic and 16 healthy dogs were lysed with proteinase K and digested with 40 U SmaI. Restriction products were separated using pulsed‐field gel electrophoresis (PFGE) with an Oxford S. aureus control and lambda‐ladder DNA concatomer markers. A dendrogram was constructed by the unweighted pair group method. All isolates showed a ≥56% similarity coefficient. Nine distinct PFGE clusters were identified, as follows: five from both atopic and healthy dogs; three from atopic dogs only; and one from healthy dogs only. Nine clusters were isolated from the nares, eight from the perineum, five from the ears and six from pyoderma lesions. There were no significant differences in the frequency of isolation from atopic or healthy skin, body sites or infected lesions for any of the clusters. Two of six healthy dogs and 18 of 20 atopic dogs with multiple isolates had closely related isolates (less than three band differences) at more than one sampling site. Isolates from pyoderma lesions were closely related to at least one mucosal isolate in 11 of 16 dogs. Staphylococcus pseudintermedius isolates appear to be heterogeneous, and colonization or infection of atopic skin was not associated with any particular strain or cluster of strains. 相似文献

The present study was conducted to investigate the relationship between waxy allelic forms and amylose in European and US rice germplasm. These allelic forms were defined according to the single-nucleotide polymorphisms (SNP) found in the leader intron 5′ splice site (G → T), exon 6 (A → C) and exon 10 (C → T). The combination of these three SNPs accounted for 89.2% of the variation in apparent amylose content in a pedigree of 85 US rice varieties and 93.8% of the variation among 279 accessions in a European germplasm collection. The allelic forms TAC and TCC were found in low amylose varieties. All varieties with intermediate levels of apparent amylose had the GCC allele. High levels of apparent amylose varieties had either the GAT and GAC allele. The sequence AGTTATA in the intron 1 distinguished the low amylose varieties from the other classes regardless of any other base changes. Intermediate amylose varieties can be distinguished from those with high apparent amylose by changes in either exon 6 or exon 10. However the simplest interpretation of the data is that the tyrosine/serine change in exon 6 is responsible for the lower levels of Granule bound starch synthase (GBSS) protein and thus lower levels of amylose in intermediate vs. high amylose verities. 相似文献

Grapefruit growers in the tropics require information about existing and new citrus cultivars with high productivity potential.
The objective of this study was to determine the growth, yield, and fruit quality performance of seven pigmented and four
white grapefruit cultivars under the dry tropic conditions of Colima, Mexico. The trees were budded on sour orange (Citrus aurantium L.) rootstock and planted at a distance of 8 × 4 m. ‘Oroblanco’ and ‘Marsh Gardner’ white-fleshed grapefruit cultivars and
‘Chandler’, a pink-fleshed pummelo, were the largest trees with the greatest height (5.0–5.6 m), canopy diameter (6.2–6.3 m),
trunk diameter (21.9–23.3 cm), and canopy volume (109–123 m3). Lower height (4.3–4.8 m) and canopy volume (73–96 m3), but with similar canopy diameter to the previously mentioned cultivars, were recorded for the remaining pigmented cultivars.
‘Chandler’ pummelo and four pigmented grapefruit cultivars (‘Shambar’, ‘Río Red’, ‘Ray Ruby’, and ‘Redblush #3’) had yearly
productions of 34.8, 34.9, 34.1, 32.7, and 30.6 ton ha−1, respectively. The most productive white grapefruit cultivar was ‘Marsh Gardner’ (30.5 ton ha−1). Grapefruit cultivars having the largest fruit size showed a higher inverse relationship between fruit weight and yield
than those with small fruit. Most genotypes had higher values of fruit weight, juice content, and maturity index than those
required by the local market. The most promising grapefruit cultivars based on their acceptable growth, yield superior to

Abstract – The downstream migration of juvenile salmonids has previously been considered predominantly passive. It has been argued that passive displacement during periods of darkness is, partially at least, a result of an inability to maintain a fixed position in the absence of visual cues. In this study, behaviour of juvenile Pacific salmonids was assessed under conditions of light and dark as they passed through an artificial channel and encountered a submerged weir. Results indicated that when light, fish formed schools and actively explored the channel. Conversely, when dark (infrared illumination only) they did not form schools, but maintained randomly distributed positions holding station against the flow. As a consequence, more fish approached and either passed, or rejected, the weir when light. The majority of fish that passed the weir did so within the first minute of each trial. Fish predominantly passed through the channel facing downstream and changed orientation prior to passing tail first over the weir crest in the presence and absence of visual cues. The orientation switch was less common when dark. This study shows, at a fine-resolution scale, that downstream movement of juvenile salmonids can be inhibited as fish exhibit alternative behaviours in the absence of visual cues. Downstream movement was not predominantly passive. Fish passage design should not be based on the assumption that downstream migration is passive. 相似文献
